“We are building regimens that are patient-centric, not product-centric,” says Barry J. Simon, MD, president and chief administrative officer of NantKwest. The company’s cell therapy strategy uses the proprietary NK-92 cell line together with an IL-15 cytokine and other immune activators. Collectively, these factors activate NK cells, dendritic cells, and T cells, allowing tumors to be attacked in a three-pronged manner. The NantKwest manufacturing process has become fully insourced and integrated. After a master cell bank is expanded serially, cells are harvested, reconstituted, and frozen for future use. After the products are sent to clinical centers, they can be readily thawed and infused into patients over an hour. Patients are treated in outpatient settings, not in hospitals. “We have transformed our cell therapy production process into a comparable but easier process than the one used to manufacture monoclonal antibodies,” asserts Simon. NantKwest has generated ample preclinical research data. To validate this data, NantKwest collaborated with Jeffrey Schlom, PhD, a senior investigator at the National Cancer Institute (NCI) and head of the NCI’s Laboratory of Tumor Immunology and Biology. The NantKwest and the NCI have been working together for several years under a Cooperative Research and Development Agreement (CRADA). “The NCI,” Simon notes, “performed a substantial amount of in vitro and in vivo research and provided valuable feedback on our products.” NantKwest currently manufactures two lead products. The first one, haNK, is a CD16, ERIL-2-enhanced NK cell line designed to be paired with a monoclonal antibody and other agents. The second product, PD-L1.t-haNK, is a human-derived allogeneic NK cell line engineered to express a CAR targeting PD-L1. Because this second product also produces intracellular IL-2 for enhanced CD16-targeted antibody-dependent cellular cytotoxicity, it can be grown without cytokine supplementation. “The IL-2 is not secreted at any detectable levels, accounting for the lack of IL-2 toxicity seen to date,” reports Simon. These two products catalyzed the development of a staged trial design, the Nant cancer vaccine. A streamlined version of this vaccine uses a tumor-conditioning agent, such as low-dose Abraxane (albumin-bound paclitaxel) or aldoxorubicin together with N-803, an IL-15 superagonist that stimulates NK- and T-cell function without inducing negative immune suppression. “We can combine this regimen with an antibody depending on what the disease is,” Simon points out. This multipronged approach neutralizes the defense of the tumor microenvironment, stimulates NK cells as the first line of defense, facilitates the NK cell to dendritic cell to T cell cross-talking, initiates the engagement with the cancer cells, and causes their lysis. “All these mechanisms,” explains Simon, “are part of a very staged protocol that we have optimized over several years and that we tested on patients with different diseases.” The use of PD-L1.t-haNK was authorized in a compassionate-use protocol for a patient with metastatic pancreatic cancer who had failed standard of care. After five infusions of PD-L1.t-haNK and N-803, the patient’s metastasis resolved completely, as shown by a CT/PET scan. “We are not saying that this is a cure,” clarifies Simon, “but it is a remarkable outcome for this patient.” This was the first patient that received the PD-L1.t-haNK as part of a chemotherapy-free combination regimen. “When we put this in context with observed responses in our other clinical trials in this and other conditions, we have enough information to file registration trials,” declares Simon. “That is what we are currently doing for non-small-cell lung, pancreatic, and triple-negative breast cancer.”

The firm currently has a $4.00 target price on the biotechnology company’s stock. Zacks Investment Research‘s price target would indicate a potential upside of 26.98% from the company’s previous close.

Das Einlizensieren, welches sehr beliebt bei Medigene ist, bietet zwar den Vorteil, dass man die Forschung schneller und kostengünstiger vorantreiben kann, beinhaltet aber auch den Nachteil, dass beteiligte Unternehmen oder Universitäten am späteren Umsatz beteiligt werden. Bei Nantkwest liegen so gut wie alle Rechte und Lizenzen beim eigenen Unternehmen.

(Nasdaq: NK), a next generation, clinical-stage immunotherapy company focused on harnessing the unique power of our immune system using natural killer (NK) cells to treat cancer and infectious diseases, today announced dosing of the first patient in a Phase 2 trial for second- and third-line Merkel cell carcinoma (MCC) in patients who are refractory to immune checkpoint inhibitors. The trial will evaluate the Company’s first-in-class, off-the-shelf CD16-targeted natural killer (haNK) cells in combination with ImmunityBio’s IL-15 superagonist N-803 and Avelumab. MCC is a rare and aggressive skin cancer that arises from uncontrolled growth of cells in the skin. Increasing in incidence, approximately 2,500 new cases are reported in the U.S. each year. Patients with metastatic or locally advanced MCC have an extremely poor prognosis, with less than 20% of patients surviving longer than five years. Typically, these patients are treated with a range of drugs, including chemotherapy, which can result in significant side effects. Although new immune therapies have the potential to improve survival, Merkel cell carcinoma is still fatal for a majority of patients who have progressed on or after treatment with a checkpoint inhibitor and represents an unmet medical need. “With over 70% of patients failing checkpoint inhibitors in solid tumors, it is vital to develop novel and effective therapeutic options,” said Patrick Soon-Shiong, M.D., chairman and CEO of NantKwest and ImmunityBio. “Checkpoint and CAR-T therapies only target the T cell, which is a small portion of effector killer cells, and often result in substantial side effects. These side effects make those therapies unusable for many patients. Our combination therapy is designed to activate the innate immune system to create an immunological memory against tumors resulting in immunogenic cell death.” “We have now established the first-in-class, cryopreserved CD16 NK-92 cell line, which can be manufactured under GMP conditions at scale. The single-arm study will involve 43 patients who failed all standard of care treatments for MCC, including checkpoint therapy (QUILT-3.063; NCT03853317). Verstehe nicht ganz was du mit Kamikazeflug und Rahm abschöpfen meinst. Es gibt Gesetzmäßigkeiten am Markt, welche man, selbst wenn man mit einem Langzeitinvestment an einer Firma gebunden bleiben möchte, ausnutzen muss oder es wenigstens versuchen sollte. Man kann später immer wieder einsteigen. Natürlich läuft man Gefahr, diesen Punkt zu verpassen, aber weder bei Medigene noch bei Nantkwest, schien mir dieser Punkt zum jeweiligen Zeitpunkt gekommen, sprich, man ist noch mindestens 1 Jahr von der Zulassung entfernt. Also kann man ganz in Ruhe das Hoch und Runter ausnutzen um noch mehr Kapital für diesen Breakthrough zu sammeln. Zudem ist Nantkwest nicht mein einziges Invest. Aber das, von dem ich mir am Meisten verspreche. Alles andere sind dividenden und renditestarke Titel.

Und hier noch mal ein kleiner Auszug über Patrick Soon Shiong, den du kleinzureden versucht hast: Soon-Shiong studierte an der University of Witwatersrand, an der University of British Columbia und an der University of California at Los Angeles Medicine & Surgery. Er ist als Hochschullehrer an der University of California in Los Angeles tätig und ist in der Krebsforschung spezialisiert. Ihm gelang unter anderem die Entdeckung von Wirkstoffen zur besseren Behandlung von Brustkrebs. 1991 gründete er das Pharmazieunternehmen Abraxis BioScience, das er 2010 an das US-amerikanische Unternehmen Celgene verkaufte, und 1997 gründete er das Pharmazieunternehmen American Pharmaceutical Partners (APP). Letzteres Unternehmen veräußerte er 2008 an das deutsche Unternehmen Fresenius Kabi. Soon-Shiong gründete auch die Stiftung Chan Soon-Shiong Family Foundation. Seit Februar 2018 ist Patrick Soon-Shiong der Eigentümer der Zeitung Los Angeles Times sowie der San Diego Union-Tribune. Soon-Shiong zahlte dafür 500 Millionen Dollar.

Ich finde es gut, wenn neue Möglichkeiten ausgelotet werden, zudem ist Nantkwest nicht allein, sondern gehört zum Nantworks Netzwerk, genau wie Nanthealth NantOmics NantBioScience NantCell •NantPharma •NantMobile •NantCapital •NantStudio Dieses Netzwerk bietet weit mehr Möglichkeiten als eine reine Pharma Firma. Das Netzwerk war teils unübersichtlich, was aber auch schon beanstandet wurde. Das Unternehmen hat einfach ganz andere Möglichkeiten als vergleichbare Konkurrenten. Und Patrick Soon Shiong hat bisher alles zu Gold verwandelt. Allerdings wird seine eigens vorgegebene Deadline mit dem ,,2020 cancer moonshot" wohl nicht einzuhalten sein. Bisher sind die Fortschritte und Ergebnisse aber vorzeigbar, weshalb man da nicht zu viel hineininterpretieren sollte. Was mich von Medigene zu Nantkwest gebracht hat, waren die oben genannten Vorteile und wegen der, wenn es zur Zulassung der Behandlung kommt, die weit günstigere Behandlungsmethode: Methode Konkurrenz: ,,Auch die zugelassenen CAR-T-Therapien von Gilead und Novartis basieren auf autologen T-Zellen. Es hat sich schon während der klinischen Phasen gezeigt, dass die Produktion dieser Zellen sowohl seitens der Qualitätskontrolle, aber auch rein logistisch große Probleme aufwirft. Nicht ohne Grund kostet eine Therapie mehrere hunderttausend US-Dollar." Methode Nantkwest: Ganz wichtig ist, dass es sich hierbei um eine universelle Therapie handelt, bei der weder Patienten- noch Spenderzellen benötigt werden. Zudem kann das Produkt in großen Mengen und hoher Qualität hergestellt und dann eingefroren werden, so dass letztlich nicht einmal ein Krankenhausaufenthalt zwingend notwendig ist, sondern die Transfusion auch beim Hausarzt durchgeführt werden kann. Das alles spart enorm an Kosten und verbessert zudem die Patientensicherheit.
